Macrocyclization methodologies allowing access to macrocycles having an endo ary ether and an endo ary-aryl bond, especially those based on an intramolecular SNAr reaction and the Suzuki-Miyaura reaction, were summarized. Total synthesis of complestatin, a bis-macrocyclic natural product, featuring these two technologies was presented.
